PompC(C) Promoter regulated by phosphorylated OmpR

This part is positively regulated by phosphorylated OmpR, a response regulator in EnvZ-OmpR two-component system. The EnvZ-OmpR system of E. coli regulates the transcriptional activity of this promoter in response to change in extracellular osmolarity. This promoter is a derivatives of the upstream of POmpC(WT)BBa_R0082 which originally comprises 3 distinct phosphorylated OmpR recognition sites, binding site 1, binding site 2 and binding site 3. OmpC(C) promoter consists of binding site 1 and additional 4 base pairs before the canonical -35 and -10 region. Figure 1. Induction of new OmpC series in high osmolarity medium at 4 hours. We characterized this promoter by attaching GFP(E0240) to the downstream and mearsuring its transcriptional activity through the GFP expression.
